...For search engines (especially google) ???

Do you mean underscores/hyphens in domain, directory, and file names?
If so, I'm pretty sure Google is able to recognize either as a word separator, so it shouldn't make much difference.
www.my-web-site.com/the_directory/the_file.html
should be the same for SEO purposes as:
www.my-web-site.com/the-directory/the-file.html
Also, you can't use underscores in domain names, only hyphens.
