In the Claim Link plug-in there is a file named email-claim-invite.txt. How, when and where is that file used?

When adding a link through 'Database -> Links -> Add' there is a text box where you can fill in an email address of somebody you want to invite to claim the link.

When you fill in this field with an email address, the template you are talking about is used to send an email to the person in question to invite them to claim the link.

Ah! Okay, that explains why I didn't see anyplace to use the file ... When adding records manually, I use Browse and select the category (catgegory list not auto generated in Database > Add form) for adding the record.
